The One Lambda™ HybriType™ NGS Hybrid Capture Assay allows for next-generation sequencing (NGS) of classical HLA, non-classical HLA, MICA/MICB and ABO blood groups (Table 1). The modular content allows for flexibility with regard to gene content, whilst extended gene coverage minimises ambiguity (providing either full gene or full exon coverage (with >85% average base coverage across intronic regions)).

The One Lambda™ HybriType™ NGS Hybrid Capture Assay utilises a probe-based design to optimise read balance and uniformity; potentially mitigating allele dropout for accurate, quality results. The streamlined workflow takes less than 5.5 hours with under 2.5 hands-on time. Sequencing data are analysed using TypeStream™ Visual NGS Analysis Software enabling rapid automated analysis and gene copy number detection. Reporting and advanced analytics are facilitated by integration within the One Lambda software ecosystem (HLA Fusion™ analysis software and HistoTrac laboratory management system).
The One Lambda™ HybriType™ NGS Hybrid Capture Assay is an excellent vehicle for genotyping known clinically-relevant HLA and ABO blood group genes, and additionally allows for the investigation of additional Major Histocompatibility Complex (MHC) genes. Non-classical HLA and MICA/MICB genes have putative roles in immune regulation, pregnancy and the outcome of haematopoietic stem cell and solid organ transplantation.
