HLA-antibody isolation
One Lambda™ MagSort™ comprises of a comprehensive panel of single HLA-antigen bound magnetic beads covering 6 classical loci – HLA-A, B, C, DR, DQ and DP. The technology enables elution of targeted HLA antibodies from complex serum, thereby facilitating focused antibody assessment without the extent of cross-reactivity which may occur in the primary serum. One Lambda™ MagSort™ can be applied to:
- Isolation of individual HLA antibodies of interest (e.g. donor specific antibody) to explore antibody properties, including through downstream cellular (e.g. flow cytometric crossmatch) and solid-phase assays (e.g. LABScreen Single Antigen & Single Antigen ExPlex).
- Enhancing eplet-level HLA-antibody specificity analysis, supported by HLA Matchmaker functionality within HLA Fusion software. Includes potential for novel eplet discovery based on unique antibody binding patterns.
- Assessment of HLA eplet immunogenicity.
- Distinguishing genuine reactivity from background reactivity and aiding in identification of weak reactivity.
- Determining polyclonal or monoclonal nature of HLA-antibody profile.
- In specific cases, indirect assessment of relative HLA-antibody quantity and binding strength.
Customisable panels
Testing is customisable with the availability of 59 antigen-specific bead sets across 6 locus-specific kits (reflecting well-documented antigens) providing flexibility to focus only on antigens of interest.
Efficient workflow
The One Lambda™ MagSort™ workflow:
- Utilises standard laboratory equipment.
- Provides immediate antigen availability as compared to alternative cell-based antibody adsorption/elution methodologies.
- Has a hands-on time of up to 45 minutes and time to completion of 3.5 hours.
The use of HLA eplets to interpret HLA-antibody data is becoming more commonplace.
